In recent years, the research field on tauroursodeoxycholic acid has been very active, and its chemical synthesis has also attracted much attention. In China, tauroursodeoxycholic acid is basically obtained through artificial extraction of bear bile, but the purity is not high and is not conducive to large-scale production. To chemically synthesize tauroursodeoxycholic acid, there are three main chemical semi-synthetic methods reported abroad:
① Use ursodeoxycholic acid (2) to react with ethyl chloroformate or pivaloyl chloride to form a mixed acid anhydride, react with taurine (5) under alkaline conditions, and then purify through an ion exchange column to obtain 1, total collection The rate is about 62%
②2 and 5 are respectively in N-ethoxycarbonyl-2-ethoxy-1,2-dihydroquinoline (EEDQ, N-ethoxy-carbonyl-ethoxy-1,2-dihydroquinoline) or cyano group under alkaline conditions Direct condensation under the action of diethyl phosphorocyanidate (DEPC) yields 1, with yields of 67% and 90% respectively. The reagents of this method are expensive;
③ After 2 and ethyl chloroformate form a mixed acid anhydride, it is condensed with p-hydroxypropiophenone to obtain the active phenolic ester of 2, which is then reacted with taurine to obtain 1. The total yield is about 64%. This method has many reaction steps. Comparing the three synthesis methods, the latter two require EEDQ and DEPC, which are expensive and cumbersome to prepare. Therefore, it is unlikely that there will be effective reagents and methods to replace bear bile in a short time.
